Een chemisch gedefinieerd, xeno- en Bloed-Free  Tradition  Medium onderhoudt verhoogde productie van kleine extracellulaire blaasjes Van mesenchymale stamcellen  Cellen

Celtherapie is getuige van een opmerkelijke verschuiving naar celvrije behandelingen op foundation van paracriene factoren, in het bijzonder naar kleine extracellulaire blaasjes (sEV), die het functionele impact van de oudercellen nabootsen. Hoewel tal van op sEV gebaseerde toepassingen zich momenteel in vergevorderde preklinische stadia bevinden, hangt hun beloofde vertaling af van het overwinnen van de fabricagehindernissen die de grootschalige productie van gezuiverde sEV met zich meebrengt. Het kweekmedium dat met de oudercellen wordt gebruikt, speelt ongetwijfeld een sleutelrol in de secretiesnelheid en inhoud van de sEV. Een essentiële vereiste is het gebruik van een serum-, xeno- en bloedvrij medium om te voldoen aan de vereisten van regelgevende instanties voor de productie van sEV’s van klinische kwaliteit . Hier hebben we Oxium TM . geëvalueerdEXO, een regelgevend medium, met betrekking tot productiecapaciteit en behoud van de kenmerken en functionaliteit van de EV en het fenotype en de levensvatbaarheid van de oudercel.

Er is een vergelijkende studie opgezet met standaard DMEM en een commercieel verkrijgbaar kweekmedium dat speciaal is ontwikkeld voor sEV-productie . Onder vergelijkbare omstandigheden vertoonde Oxium TM EXO een drievoudige toename van de sEV-secretie, met een verrijking van deeltjes tussen 51 en 200 nm. Deze resultaten werden verkregen door directe kwantificering van het geconditioneerde medium om interferentie en variabiliteit van de isolatiemethode te vermijden en werden vergeleken met de twee kweekmedia die werden geëvalueerd. De hogere opbrengst die werd verkregen was constant met verschillende oogsttijdstippen (2, four en 6 dagen) en verschillende celbronnen, waaronder navelstreng-, menstruatiebloed-afgeleide mesenchymale stromale cellen en fibroblasten. Bovendien zijn het stamcelfenotype enlevensvatbaarheid van de oudercel bleef ongewijzigd.

Bovendien vertoonde Oxium TM EXO-sEV een vergelijkbaar expressiepatroon van de vesiculaire markers CD63, CD9 en CD81, met betrekking tot sEV afgeleid van de andere aandoeningen. De  in vitro  internalisatie-assays in verschillende doelceltypen en het farmacokinetische profiel van intraperitoneaal toegediende sEV  in vivo  gaven aan dat de hogere EV-productiesnelheid geen invloed had op de opnamekinetiek of de systemische biodistributie bij gezonde muizen. Samenvattend, het Oxium TM EXO-medium ondersteunt een efficiënte en robuuste productie van grote hoeveelheden sEV , waarbij de klassieke functionele eigenschappen van internalisatie in acceptordoelcellen en biologische distributie  in vivo worden behouden, het leveren van de hoeveelheid en kwaliteit van EV’s voor de ontwikkeling van celvrije therapieën.

Oprichting en lange termijn  cultuur  van de muis mamma-stam  cel  organoids en borsttumor organoids

Mammaire stamcellen (MaSCs) bijdragen tot borstepitheel ontwikkeling en homeostase. Ze zijn voorgesteld als cellen van oorsprong voor borstkanker. Hier beschrijven we een organoïde cultuurprotocol voor  ex vivo  uitbreiding van MaSC’s van muisweefsels. Deze organoïden behouden de zelfvernieuwing van klier-reconstituerende MaSC’s en kunnen worden gebruikt om tumorigenese te modelleren door patiëntrelevante kankerfactoren en mutaties te introduceren.

Vergelijkbare organoïde cultuur kan worden gebruikt voor langdurige expansie van luminale stam-/voorlopercellen van normale klieren en tumor-initiërende cellen van borsttumoren . Raadpleeg Christin et al. voor volledige particulars over het gebruik en de uitvoering van dit protocol. (2020) en Zhang et al. (2016).

Poly (amino ester) gebaseerde polymeren Gene and Drug Supply Methods en verdere toepassing richting  Cell  Tradition  System

Verschillende synthetische polymeren op foundation van poly(amino-ester) (PAE) worden gesuggereerd als kandidaten voor gen- en medicijnafgifte vanwege hun pH-responsiviteit, wat bijdraagt ​​aan efficiënte leveringsprestaties. PAE-gebaseerde pH-responsieve polymeren zijn meer biologisch afbreekbaar en hydrofiel dan andere soorten pH-responsieve polymeren. De functionaliteit van op PAE gebaseerde polymeren kan worden versterkt door verschillende chemische modificaties te gebruiken om de efficiëntie van gen- en medicijnafgifte te verbeteren. Bovendien worden op PAE gebaseerde polymeren op veel manieren gebruikt in het biomedische veld, zoals in transdermale toediening en stamcelkweeksystemen.

De effecten van genotypen en media samenstelling op callogenesis, regeneratie en  cel-  suspensie  kweken  van kamille (  Matricaria chamomilla  L)

Achtergrond:  Kamille is een belangrijk kruid dat veel wordt gebruikt voor medicinale doeleinden. De multitherapeutische, cosmetische en voedingswaarden zijn vastgesteld door jarenlang traditioneel en wetenschappelijk gebruik en onderzoek. Een toenemend gebruik van geneeskrachtige planten vereist zowel rationeel gebruik als duurzame productie van dergelijke genetische bronnen. In vitro microvermeerdering van planten biedt unieke kansen voor duurzame productie van geneeskrachtige kruiden, hun hergroei en conservering. De huidige studie was gericht op het onderzoeken van de effecten van verschillende explantaten, combinaties van plantengroeiregulatoren (PGR’s) en mediatype op callogenese, in vitro regeneratie en celsuspensie van zes kamille-genotypen om de duurzame productie ervan te verbeteren.

Methoden:  De scheut-, zijspruit- en blad-afgeleide explantaten van zes kamille-genotypen, waaronder Isfahan, Shiraz, Kazeron, Goral, Sharokashari en Presso, werden gebruikt voor directe en indirecte regeneratie. Voor indirecte regeneratie werden verschillende doses NAA en kinetine gebruikt om calli te induceren die werden gekweekt op MS-media die PGR’s bevatten voor directe en indirecte regeneratie. Later werd celsuspensie vastgesteld en werd morfologische karakterisering van met CrO 3  gekleurde cellen uitgevoerd met behulp van microscopie.

Resultaten en discussie:  Onze bevindingen toonden aan dat het hoogste calluspercentage en callusvolume werden waargenomen van laterale spruiten en scheuten van genotype Isfahan op MS-medium dat 1 mg/L NAA en 1 mg/L kinetine bevat. De in vitro regeneratie bleek genotype-afhankelijk te zijn, terwijl 77% en 77,5% het hoogste proportion was voor respectievelijk indirecte en directe regeneratie. Bovendien werden het maximale aantal scheuten (twee scheuten/explantaat) en scheutlengte (2,22 cm) ook waargenomen in het Isfahan-genotype. Celsuspensiekweek vertoonde het hoogste versgewicht (18,59 g) en drooggewicht (1,707 g) met 0,75 g entmateriaal van de callus afkomstig van zijspruiten gekweekt op MS-medium. Microscopie van CrO 3 gekleurde cellen werden gedurende 27 dagen op elke 3e dag gedragen, wat in de begindagen grotere en sponsachtigere cellen onthulde in vergelijking met de laatste dagen toen het aantal cellen groter was maar de celgrootte kleiner was.

Conclusie:  De callogenese, organogenese en celsuspensiecultuur van kamille kan afhankelijk zijn van het genotype. Daarom is optimalisatie van media-ingrediënten en kweekomstandigheden van het grootste belang voor het bedenken van een op weefselkweek gebaseerde conserveringsstrategie van elk kamille-genotype en de productie van secundaire metabolieten.
